Flow Cytometer in EECF Flow Cytometer - EECF

The lab has a single Flow Cytometer system.

Users are required to take the “Introduction to Flow Cytometry lecture series” provided by the by UWCCC Flow Lab (cost is $85 for UW-Madison faculty and students; NOTE: cost is set by the UWCCC and may change). This is a 3-part class (90 minutes each) that provides a great overview of flow cytometry and instrumentation.

The lab has multiple GC (Gas Chromatography) systems for the analysis of organic compounds in water, solvent, or gas.

  • Manufacturer (all): Shimadzu Scientific.

(1) GC #3 – w/ ECD (Electron Capture Detector) and user provided column(s)

(2) GC #4 (for gas analysis) w/ both FID (Flame Ionization Detector) and TCD (Thermo Conductivity Detector); installed with system-specific Packed Columns

(3) GC #5 – w/ FID (Flame Ionization Detector) and a variety of columns to choose from [Stabilwax-DA (15m), Stabilwax-DA (30m), Restek RTX-624 (30m)]

Samples must be extracted into a solvent (unless using the Stabilwax column on GC5 for fatty acid analysis – can analyze water samples). A set of calibration standards should be prepared in the same solvent.

GC-MS

The lab has a single GC-MS (Gas Chromatography-Mass Spectrometry) system for the analysis of organics.

  • Model: GC-2020 with QP2010S Mass Spectrometer detector
  • Column: Restek RXI-5MS (30m, 0.25mm ID, 0.25um)

Samples must be extracted into a solvent. A set of calibration standards should be prepared in the same solvent.

HPLC

The lab has a single HPLC (High Performance Liquid Chromatography) system for the analysis of a variety of analytes from anions to sugars.

  • Detectors: Diode Array (UV-Vis); Refractive Index (RI); Fluorescence
  • Column(s): Restek Ultra Aqueous C18 (4.6 x 150mm, 5um); Bio-Rad Aminex HPX-87H (300 x 7.8 mm, 9um)

All samples must be filtered with a 0.45um filter and in 1.7-2.0mL autosampler vials. A set of calibration standards should be prepared under the same conditions as the samples.

ICP

The lab has a single ICP-OES (Inductively Coupled Plasma – Optical Emission Spectrometry) system for the analysis of Metals.

Liquid samples must be filtered using a 0.45um filter, adjusted with concentrated Nitric Acid (HNO3) to pH <2, and brought to the lab in 15mL disposable centrifuge tubes (like THESE). A set of calibration standards should be prepared under the same conditions (pH <2 with HNO3, 15mL tubes).

The lab has a single epi Fluorescent microscope for both fluorescence and bright field microscopy.
